2019年1月7日 讯 /生物谷BIOON/ --诱发癌症有很多遗传原因,比如有些突变会遗传自父母,而其它则是后天获得性的突变,比如因外界因素或DNA复制的错误导致等,大规模的基因组测序在识别因体细胞突变所诱发的癌症上取得了一系列研究成果,但这种技术却无法有效识别因遗传性突变所诱发的癌症,而且识别这些遗传突变的主要来源仍然是基于家族性的研究。图片来源:iran-daily.com近日,一项刊登在国际杂志Nature Communications上的研究报告中,来自巴塞罗那基因组研究中心的科学家们通过研究开发了一种新型的统计学方法,其能够从肿瘤测序数据中鉴别出癌症易感性基因;该方法使用了一种古老的想法,即癌基因通常需要“两次击打”(two hits)才能够致癌,研究者表示,这种方法能帮助他们从当前癌症基因组数据库中系统性地鉴别出相关的基因。研究者将这种统计学方法命名为“ALFRED”,目前该技术已经鉴别出了13个候选的癌症易感性基因,其中10个基因都是此前并未发现的;Ben Lehner博士说道,我们对30种不同类型的肿瘤患者进行研究,将这种方法应用于1万多名患者的基因组序列中,最终鉴别出了已知的和一些新型的可能性癌症易感基因,这些基因有可能会诱发一定的患癌风险。研究者指出,新型的癌症易感基因或许在多种类型癌症的发生上扮演着关键角色,比如,其与14%的卵巢肿瘤、1%的乳腺癌等癌症直接...

冬季流感严重危害人类健康,甚至会威胁生命,我们究竟有什么办法预防流感呢?目前,美国密歇根大学 Betsy Foxman 和同事最新研究表明,呼吸道微生物群可能与流感易感性密切相关。这项研究报告发表在 1 月 9 日出版的《美国公共科学图书馆 · 综合》杂志上。流感病毒是流感病原体,主要侵袭目标是人体呼吸道,感染上皮细胞。鼻子和喉咙的上皮细胞被复杂的细菌群落包围,因此 Betsy Foxman 和同事推测认为,这些呼吸道微生物群可能与病毒相互作用,并在人体对流感预防方面充分发挥作用。在这项研究中,研究人员对 2012—2014 年登记的 144 个尼加拉瓜家庭进行了分析,这些家庭中有一位成员被确认患有流感,包括成年人和儿童的所有家庭成员都接受为期两个星期的监测。研究人员对每个家庭进行 5 次回访信息统计,对所有家庭成员的呼吸道微生物取样,并对监控测试者进行症状日志记录。研究者使用统计模型,对每次测试的家庭成员呼吸道微生物群分类为两个不同种群类型,尽管大约一半的测试者在研究测试过程中体内微生物群类型发生了变化。接下来,研究人员将参与者微生物类型与他们流感易感性进行了对比分析。结果发现,跨年龄组的一种呼吸道微生物类型显示出对流感的易感性降低,这种微生物类型在婴儿和儿童群体中并不常见,而且与年龄较大的青少年和成年人相比,其稳定性较低。虽然微生物种群和流感易感性之间的联系在这项研究...

日前,《分子植物》(Molecular Plant)在线发表了中国农业科学院油料作物研究所王汉中院士团队最新成果。该团队在国际上首次成功克隆了农作物种子性状的第一个细胞质调控基因 orf188,并揭示了该基因调控油菜种子高含油量的作用机制。该成果为油菜高含油量育种提供了新途径,为育种过程中杂交母本的选择提供了理论支撑,对于农作物不同类型细胞质的应用具有重要的指导意义。含油量是油菜产油量最重要的构成因子之一,随着现代育种对含油量潜力的不断挖掘,提高品种的含油量越来越困难。该团队在油菜含油量遗传研究的基础上,从细胞质效应着手对含油量母体调控新机制研究取得突破,筛选到两个对含油量具有极显著细胞质正负效应的品种,鉴定出特异基因 orf188 控制细胞质的含油量正效应,过表达该基因可以大幅度提升含油量。研究表明,orf188 为 nap(nap-like)型油菜品种所特有基因,是一个新近进化形成的嵌合基因。该团队十余年来围绕油菜种子性状的母体调控进行系统研究,此次对细胞质基因调控种子含油量机理的揭示,是该团队继发现角果皮发育调控种子粒重和含油量之后发现的又一条母体调控新途径,进一步丰富和完善了种子性状的母体调控理论,也为其他农作物性状细胞质效应的解析提供了借鉴。相关论文信息:DOI:10.1016/j.molp.2019.01.012来源:中国科学报转载:生物360
